Uses

6-Chloro-4-hydroxy-2-methylquinazoline is a quinazolinone/quinazoline derivative.

A magnetic stirrer was charged into a two-necked round-bottomed flask, evacuated and displaced three times with nitrogen. An aqueous solution (3 mL) of 2 wt% TPGS-750-M in 2-bromo-5-chlorobenzoic acid (1, 0.5 mmol) and acetamidine hydrochloride (2, 0.75 mmol) was added under nitrogen protection. After stirring for 10 min, Cs2CO3 (1 mmol, 326 mg) was added to the flask. 15 min later, CuI (0.1 mmol, 19 mg) was added. The reaction mixture was stirred at the indicated temperature (see Table 3 in the text) for 12 h under nitrogen atmosphere. Upon completion of the reaction, the mixture was extracted with ethyl acetate (1 mL) and concentrated under reduced pressure. The residue was purified by silica gel column chromatography using petroleum ether/ethyl acetate (3:1 to 1:1) as eluent to afford 6-chloro-2-methylquinazolin-4-one.
