6-Hydroxy-7-methoxyquinazolin-4-one (1.92 g, 10 mmol) was placed in acetic anhydride (20 mL) and pyridine (4 mL) was added slowly with stirring. The reaction mixture was heated to 100 °C and kept for 4 hours. After the reaction was completed, it was cooled to room temperature and ice water was added to quench the reaction. The white solid that precipitated was the intermediate 7-methoxy-4-oxo-3,4-dihydroquinazolin-6-ol acetate (M-1, 2.32 g, 99% yield).
