Uses

(S)-(+)-1-Amino-2-propanol may be used in the preparation of S-(+)-1-(2,3-naphthalimido)-2-propanol.

General Description

(S)-(+)-1-Amino-2-propanol is a chiral amino alcohol.

Synthesis

Step (1): dissolve sodium tert-butoxide (230g, 2.39mol) in tetrahydrofuran (2L), slowly add trifluoroacetamide (226g, 2mol) under the ice bath, stirring for 30min; then add S-epoxypropane (128g, 2.2mol) under the ice bath, naturally warmed to room temperature, and continue to stir the reaction for 10h, and then stirred the reaction for another 2h at 35??; after the reaction After the reaction, 2N (equivalent concentration) of hydrochloric acid was added dropwise to the system for neutralization, and then 1L of water was added, static layering, the aqueous phase was extracted with dichloromethane, the organic phases were combined, dried with anhydrous sodium sulfate, and concentrated under pressure to obtain a total of 318g of intermediate product I, with a yield of 93%. Step (2): dissolve the intermediate product I (310g, 1 .81mol) obtained in step (1) in methanol (1 .5L), add 200mL of water and potassium carbonate (550g, 4mol), stirring at room temperature for 6h, filtration, concentration to dryness, the product obtained was dissolved in dichloromethane, filtration, insoluble matter was removed, anhydrous sodium sulfate was dried, concentration, distillation under reduced pressure, the target product S 1-amino-2-propanol totaling 122 g in 90% yield.
