Synthesis

Preparation of dexamethasone sodium phosphate for injection. Synthesis: Using dexamethasone acetate epoxide as the starting material, steps include a ring-opening reaction (HF/DMF, -10??C), recrystallization (methanol), base-catalyzed hydrolysis (Na2CO3/methanol, 20??C), esterification with pyrophosphoryl chloride (THF), and neutralization/salt formation (NaOH/methanol, 20??C) to yield a dexamethasone sodium phosphate solution, which is recrystallized again to give crystals. The crystals are dried and ball-milled, then mixed with glucose, disodium hydrogen phosphate, and sodium dihydrogen phosphate. This mixture is dissolved in water for injection, ethanol and activated carbon are added, the solution is kept warm, and then filtered.
