Description
Phenobarbital-d5 (ethyl-d5) is an analytical reference material intended for use as an internal standard for the quantification of phenobarbital by GC- or LC-MS. Phenobarbital is characterized as a barbiturate. Phenobarbital-d5 (ethyl-d5) is regulated as a Schedule IV compound in the United States. This product is intended for research and forensic applications.

Definition
ChEBI: Phenobarbital-d5 is a pyrimidone and a deuterated compound.
Synthesis
Phenobarbital-D5 was prepared as follows: 1.0 g (0.0166 mol) of urea and 1.0 g (0.0416 mol) of sodium hydride were added to a 50 ml round bottom flask equipped with a stir bar, a reflux condenser and a rubber septum under nitrogen. While stirring, 3.4 g (0.0126 mol) of diethyl ethyl-D5-phenylmalonate in 40 ml of anhydrous ethanol was slowly added through a cannula. The mixture was refluxed for 24 hours and then added to 50 ml of 0.1 NH2SO and 25 ml of crushed ice. After extraction with ether, drying and evaporation of the solvent to a small volume, phenobarbital-D5 was obtained by column chromatography (silica gel; E.M. reagent, 0.5-0.2 mm mesh, 2.5x28 cm) by addition of 200 ml of anhydrous ether. The ether was evaporated to give 2.0 g (65%) of pure phenobarbital-D5.
