General procedure for the synthesis of 2-chloro-5-iodobenzotrifluoride from 2-trifluoromethyl-4-iodophenol: 4-iodo-2-trifluoromethylphenol (10 mmol) obtained in 1.1 was dissolved in 20 mL of N,N-dimethylformamide (DMF). Subsequently, phosphorous trichloride (10.5 mmol) was added, and the reaction system was placed in a microwave reactor, heated to 130 °C, and kept for 1 hour. After the reaction was completed, the system was cooled to room temperature, 30 mL of water was added and stirred for 10 minutes. The reaction mixture was extracted with 30 mL of dichloromethane and the organic phase was dried with anhydrous sodium sulfate. After filtration, the filtrate was concentrated under reduced pressure. Purification by fast column chromatography using petroleum ether as eluent gave 2.1 g of light yellow solid 1-chloro-4-iodo-2-trifluoromethylbenzene in 69% yield.
