Paederosidic acid Usage And Synthesis

Chemical Properties

Yellow powder, soluble in methanol, ethanol, DMSO and other organic solvents, derived from Paederia scandense.

Uses

Steinylic acid has analgesic and anti-inflammatory effects.It is used for content determination/identification/pharmacological experiments, etc.

Synthesis

The method of extracting and isolating chickweed glycosides from the agarwood tree has the following steps: (1) The dried and crushed stem powder (10 kg) of agarwood tree with particle size of 50-200 mesh was extracted three times (the first time for 5 days, the second time for 7 days, and the third time for 8 days) with an aqueous ethanol solution (10 kg) with a volumetric concentration of 75%, and the extracts were combined, and a brown viscous ethanol extract (437 g) was obtained by concentrating the extracts under reduced pressure. The ethanol extract obtained in step was homogeneously dispersed in 1000 mL of distilled water, and sequentially extracted three times with equal volumes of petroleum ether (1000 mL) and ethyl acetate (1000 mL), the extracts were combined, and the organic phase portion (30 g of petroleum ether portion and 20 g of ethyl acetate portion) was obtained by concentration under reduced pressure, and the remaining aqueous phase portion (240 g) was further dispersed in 500 mL of distilled water, stirred homogeneously, and then subjected to macroporous concentration. The remaining aqueous phase portion (240 g) was then dispersed in 500 mL of distilled water, stirred well and subjected to macroporous resin chromatography. (iii) The crude chickweed vine sapogenins obtained in step was dissolved in methanol to a concentration of 0.5 g/mL, and then subjected to silica gel column chromatography, with the mesh number of silica gel being 250 mesh, and the dosage of silica gel being 8 g. The gradient elution was carried out by using chloroform-methanol in accordance with the gradient of 100:0~0:100 (V/V), and the streams were collected in every 50 mL, concentrated, and then detected by TLC, and then combined with the same streams to combine the streams, and then the chickweed vine sapogenins with the purity of 90% (the streams of 90%) were separated into two streams. The 90% fraction (the fraction eluted from chloroform:methanol=1:2) was dried to obtain 95% purity of chickweed vine sapogenetic acid refined product (1.65 g).

Physiological effects

Paederosidic acid has good peripheral analgesic effect and central nerve analgesic effect, and also has good anti-inflammatory effect.
