Uses
Agonist of the orphan G protein-coupled receptor GPR35: a potent activator of extracellular signal-regulated kinase and β-arrestin2 with antinociceptive activity. Used as an inhibitor in the real-time fluorescence enzymatic characterization study of specialized human DNA polymerases.
Uses
Pamoic acid is used in preparation of repository derivatives of medicinal agents, used in pharmacology to increase the solubility of a drug in water.

General Description

Biological Activity
(1)Pamoic acid can inhibit Cas9 endonuclease activity. It effectively blocks bacterial or mammalian genome editing mediated by SpyCas9, SauCas9 or BE4 base editors at multiple endogenous gene loci. In addition, pamoic acid displays substrate competitive and PAM binding site occupancy mechanisms that selectively inhibit linear DNA cleavage compared to Cas9 cleavage of superhelical DNA. Importantly, pamoic acid pharmacologically modulates Cas9-mediated genome editing in mice.
(2)GPR35 activation by Pamoic acid may increase the phosphorylation of ERK1/2, which in turn initiates an anti-inflammatory signal by suppressing NF-κB-dependent inflammatory genes.
Safety Profile
Poison by intraperitoneal route. An eye irritant. When heated to decomposition it emits acrid smoke and irritating fumes.
