5-Aminomethyl-2-chloropyridine is a reagent that is used in the synthesis of macamides and analogs that have FAAH (fatty acid amide hydrolase) inhibitory activity.

General procedure for the synthesis of 5-aminomethyl-2-chloropyridine from the compound (CAS:1080018-12-7) (Example 5): preparation of (2-chloropyridin-5-yl)methylamine (4). To 0.77 g (1.66 mmol) of 1,3,5-tris[(2-chloropyridin-5-yl)methyl]-1,3,5-perhydrotriazine (II-4) were sequentially added 0.40 g of methanol and 1.83 g of concentrated hydrochloric acid, and the resultant mixture was stirred for 6 hr. at 75 to 80° C. The reaction was carried out with the aid of a liquid chromatograph. After completion of the reaction, the reaction mixture was diluted with chloroform and adjusted to basicity by addition of 28% aqueous sodium hydroxide. The chloroform layer was separated and concentrated to give 0.68 g (yield: 95%) of the target product (2-chloropyridin-5-yl)methylamine (III-1).
