Tat-Beclin 1 Peptide Usage And Synthesis
Uses
Tat-beclin 1, a peptide derived from a region of the autophagy protein (beclin 1), is a potent inducer of autophagy and interacts with negative regulator of autophagy, GAPR-1 (GLIPR2). Tat-beclin 1 decreases the accumulation of polyglutamine expansion protein aggregates and the replication of several pathogens (including HIV-1) in vitro, and reduces mortality in mice infected with chikungunya (CHIKV) or West Nile virus (WNV)[1].
General Description
The TAT-BECLIN-1 (autophagy activator) peptide is derived from a region of the autophagy protein, beclin 1, attached to the HIV-1 Tat protein transduction domain. The peptide includes 11 amino acids at the N terminus derived from the Tat protein transduction domain, 18 amino acids (267–284 aa) from beclin 1 at the C terminus and a GG linker.
Biochem/physiol Actions
The TAT-BECLIN-1 (autophagy activator) peptide reduces the accumulation of polyglutamine aggregates and pathogen replication in vitro. This peptide is also found to reduce the mortality of mice infected by different viruses. The TAT-BECLIN-1 peptide is predicted to stimulate cell death via autophagy.
in vivo
Tat-beclin 1 (15 mg/kg; i.p.; daily; beginning 1 day post-infection for 20 days) can induce autophagy in peripheral tissues in adult mice as well as in the central nervous system of neonatal mice (6-week-old GFP-LC3 mice)[1].
