To a reaction flask was added 200 mL of methanol, 52.08 g of 2-amino-3-nitro-6-chloropyridine (compound 1), 37.54 g of p-fluorobenzylamine and 50.0 mL of triethylamine. The reaction mixture was heated to 80 °C and kept for 10 hours. After completion of the reaction, 10 times the volume of water was added to the reaction solution and a solid was precipitated, which was filtered and dried to give 76.47 g of 2-amino-3-nitro-6-(4-fluorobenzylamino)pyridine (Compound 3) in 97.2% yield and 99.62% purity (as determined by HPLC).
