Uses

6-Nitroisoindoline-1-one is a heterocyclic derivative that can be used as a pharmaceutical intermediate. It exists in a variety of natural products and pharmaceutical molecules and has extremely high biological activity.

General procedure for the synthesis of 6-nitro-isoindolin-1-one from methyl 2-(bromomethyl)-4-nitrobenzoate: Methyl 2-(bromomethyl)-4-nitrobenzoate (250 mg, 0.92 mmol) was added to a methanolic solution of saturated ammonia (5 mL) and stirred for 2 hr at room temperature, and completion of the reaction was monitored by thin layer chromatography (TLC). The solvent was removed by distillation under reduced pressure and ethyl acetate (7 mL) was added to the residue and sonicated for 3 min, followed by placement in a refrigerator at -20 °C for 3 min to promote crystallization. After precipitating a large amount of solid, the product was collected by filtration and dried under high vacuum to give 150 mg of a pale yellow solid in 92% yield.
