Sulforhodamine B Usage And Synthesis
Chemical Properties
This substance is a deep red powder. It is easily soluble in water, forming a bluish-red solution with yellow fluorescence. When sodium hydroxide solution is added, it also appears bluish-red. Additionally, it is soluble in ethanol. In concentrated sulfuric acid, it turns orange-yellow and red after dilution.
Uses
Sulforhodamine B sodium salt has been used:
- for relative total cell mass quantification
- to investigate the viscosity of coacervates
- as a molecular rotor for microviscosity determination in aerosol droplets
Uses

Application
Sulforhodamine B (SRB) is known for use in SRB assay for the determination of in vitro cytotoxicity through staining and the observation of live cells, proliferation and drug screening.
It has been used in the SRB assay to evaluate the effect of mouthwashes on protein content in primary cultures of human fibroblasts.
It is also suitable as a microscopic tracer for the assessment of blood-brain barrier integrity by post mortem inspection of frozen sections. SRB is useful as a caries detector dye with subsequent inspection of histological material.
Preparation
4-Formylbenzene-1,3-disulfonic acid and 3-(Diethylamino)phenol condensation, the products with sulfuric acid dehydration, and ferric chloride oxidation, and finally into sodium salt.

General Description
Sulforhodamine B (SRB) is a bright-pink aminoxanthene dye. The sulforhodamine B (SRB) assay is extensively used for in vitro cytotoxicity screening. SRB binds to the protein constituents of the cells in a stoichiometric manner. Thus, the amount of dye extracted from stained cells is directly proportional to the cell mass. SRB can also be used for cell density determination.
