SteMRegenin 1 Usage And Synthesis
Description
SR1 (1227633-49-9)?extends the pluripotency of hematopoietic stem cells by antagonizing the arylhydrocarbon receptor (AhR), IC50?= 127?nM.1?The effect is dose-dependent and reversible. Displays a biphasic effect on CD34-negative cells with low does (1 μM) enhancing proliferation and high does (>5 μM) repressing proliferation.2?May be used to generate progenitor T cells3?from HSPCs as well as NK cells for immuno-oncology studies4.
Application
StemRegenin 1 can also be used to treat infectious diseases such as HIV/AIDS, malaria, and tuberculosis. The mechanism by which StemRegenin 1 stimulates the production of new red blood cells is not well understood; however, it appears to work through activation of JAK1. This protein inhibits the polymerase chain reaction (PCR), which is an enzyme that increases the rate at which DNA copies are made from a template molecule. Inhibitors of this enzyme have been shown to prevent infection with Human Immunodeficiency Virus (HIV) in vitro.
General Description
A cell-permeable purine compound that acts as a high affinity AhR antagonist (IC50 = 127 nM against 3 nM TCDD-induced luciferase reporter activity in HepG2 cultures), but not a panel of 61 kinases. AhR expression and function knockouts by shRNAs, CH-223191 (Cat. No. 182705), or SR1 treatment all result in enhanced and sustained CD34+ population in long-term proliferating/expanding cultures of cord blood-derived HSCs (hematopoietic setem cells) in serum-free media containing Tpo (Cat. No. 605218), SCF (Cat. No. 569600), Flt3 Ligand (Cat. No. 343010), and IL-6 (Cat. No. 407652) in a reversible manner. SR1 is reactive toward human, monkey, and canine, but not rat or murine, species.
Uses
StemRegenin 1 is a plant-derived stem cell factor that is potent inducers of hematopoietic stem cells. It has been shown to stimulate the differentiation and proliferation of primary cells, such as human umbilical cord blood cells and bone marrow cells, into mature erythrocytes and leukocytes.
