Synthesis

Compounds 1-6 (7.2 g, 30 mmol) were used as starting materials and dissolved in 100 mL of n-pentanol. The temperature of the reaction system was maintained at 110 °C and 6.9 g of sodium metal was added in batches. After the sodium metal was completely reacted, the reaction was continued with stirring for 1 hour, followed by cooling to room temperature. 100mL of water was added to the reaction mixture and the aqueous phase was separated. The organic phase was washed three times with 450mL of 2N hydrochloric acid solution. The solvent was removed by rotary evaporator and the resulting residue was dissolved in 100 mL of aqueous 2N NaOH solution and subsequently extracted three times with 300 mL of dichloromethane. The organic phases were combined, washed twice with saturated aqueous sodium chloride solution and dried with anhydrous sodium sulfate. To the dried organic phase was added 100 mL of 2N hydrochloric acid, and the solvent was removed by concentration via rotary evaporation to give the final target product 3,3-dimethylazetidine hydrochloride (Compounds 1-7, 0.9 g, 24.7% yield) as a white solid.
