General procedure for the synthesis of 6-formyl-2-pyridinecarbonitrile from 6-hydroxymethyl-2-cyanopyridine: P4: For alternative synthesis see literature [36-38]. The reaction was carried out under nitrogen protection. Selenium dioxide (SeO2, 386 mg, 3.48 mmol) and 6-hydroxymethyl-2-pyridinecarbonitrile (933 mg, 6.96 mmol) were dissolved in dioxane (30 mL, without drying and degassing) and the reaction was stirred at 110 °C (oil bath temperature) for 24 h. The reaction mixture turned into a dark yellow solution accompanied by a black selenium precipitate. Upon completion of the reaction, the mixture was filtered through diatomaceous earth to remove the selenium precipitate and the diatomaceous earth was washed with dichloromethane (CH2Cl2). Purification was carried out by silica gel column chromatography (10 g silica gel, eluent CH2Cl2) and the light yellow eluate containing the product was collected to give the white solid product 6-formyl-2-pyridinecarbonitrile (692 mg, 5.24 mmol, 75% yield; molecular formula C7H4N2O, molecular weight 132.12).
