Sacubitril Impurity 7 Usage And Synthesis

Uses

Desethyl Sacubitril is an impurity in the synthesis of Sacubitril (S080895), an antihypertensive drug used in combination with valsartan. The combination drug, valsartan/sacubitril, known during trials as LCZ696 and marketed under the brand name, Entresto, is a treatment for heart failure.

Biological Activity

LBQ657 is the active metabolite of sacubitril. It is an inhibitor of the enkephalinase neprilysin (neutral endopeptidase, NEP) formed by the action of esterases on its prodrug sacubitril (AHU377), which is half of the heart failure combination drug Entresto, along with the angiotensin-receptor blocker Valsartan. LBQ657 prevents neprilysinμs degradation of atrial and brain natriuretic peptide, two blood pressure lowering peptides.
