Hyaluronidase Usage And Synthesis
Chemical Properties
Vitaminase is an enzyme extracted from mammalian testes that can hydrolyze glassy mucopolysaccharides, it is a white or slightly yellow powder, soluble in water, insoluble in ethanol, acetone or ether. Odorless. Stability is good, 420C continuous heating for 60 min without loss of vitality, 1000C continuous heating for 5 min can retain 80% of the vitality. It is easy to be inactivated in low concentration of aqueous solution, and can be protected by gelatin or gum arabic to prevent inactivation.Fe2+ and Cu2+ ions have reversible inhibition on the enzyme, while Pb2+, Hg2+, Ni2+ and other ions have no significant effect on the enzyme activity. The inhibitory effect of chondroitin B sulfate (dermatophyte), heparin sulfate, keratin sulfate, sodium heparin, and high concentrations of vitric acid on vitreous acid enzyme was reversed in the presence of sodium chloride (0.15 mol/L) or fisetin sulfate. Vitaminase Injection is a sterile, freeze-dried product of vitaminase plus a suitable excipient as a white or off-white lyophilized mass or powder. The aqueous solution is unstable and needs to be prepared before clinical use.
Uses
Pharmaceutic aid (diffusing agent in s.c. injections).
brand name
Diffusin (Ortho Pharmaceutical); Enzodase (Bristol-Myers Squibb); Hyazyme (Abbott); Vitrase (Biozyme Laboratories Ltd., UK); Wydase (Wyeth-Ayerst);Hylase.
Purification Methods
Hyaluronidase is purified by chromatography on DEAE-cellulose prior to use. [Distler & Jourdain J Biol Chem 248 6772 1973.]
