Description

B355252 is a neuroprotective agent. It potentiates NGF-induced neurite outgrowth in NS-1 cells (EC₅₀ = ~1 μM). B355252 inhibits glutamate-induced excitotoxicity in HT-22 cells in a concentration-dependent manner and inhibits glutamate-induced decreases in GSH levels and increases in Bax levels, intracellular accumulation of calcium, and production of reactive oxygen species (ROS) in HT-22 cells when used at a concentration of 8 μM. It also inhibits decreases in cell viability, increases in ROS production, and mitochondrial membrane depolarization induced by 6-hydroxydopamine (6-OHDA; ) in HT-22 cells, an in vitro model of Parkinson’s disease.

Uses

B355252, a phenoxy thiophene sulfonamide small molecule, is a potent NGF receptor agonist. B355252 potentiates NGF-induced neurite outgrowth. B355252 protects ischemic neurons from neuronal loss by attenuating DNA damage, reducing ROS production and the LDH level, and preventing neuronal apoptosis. B355252 has anti-apoptotic effects in glutamate-induced excitotoxicity, as well as in a murine hippocampal cell line (HT22) model of Parkinson disease (PD)[1].

in vivo

B-355252 (0.125 mg/kg; ip; daily; 3 days) significantly attenuates the infarct volume and protects post-stroke neuronal loss in adult male Sprague–Dawley rats with cerebral ischemia^[1].
