General procedure for the synthesis of 1H-pyrazolo[3,4-c]pyridin-3-amine using 3-fluoro-4-cyanopyridine as starting material: 3-fluoro-4-cyanopyridine (0.50 g, 4.10 mmol) was dissolved in ethanol (8 mL), followed by addition of hydrazine hydrate (0.31 g, 6.1 mmol). The reaction mixture was stirred at 70 °C for 16 hours. Upon completion of the reaction, the mixture was cooled to room temperature and subsequently concentrated and dried under vacuum to afford the target product 1H-pyrazolo[3,4-c]pyridin-3-amine (0.66 g, 100% yield). The product was characterized by LC-MS (Method 2B): retention time (Rt) = 0.51 min, mass spectrum (ESI positive ion mode): m/z = 135 [M + H]+.
