Step A: To a solution of N,N-dimethylformamide (DMF, 5 mL) of 2-amino-3-bromobenzoic acid (0.5 g, 2.3 mmol) was added hydroxybenzotriazole (HOBt, 0.46 g, 3.0 mmol), 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC-HCl, 0.53 g, 2.8 mmol), ammonium chloride (0.5 g, 9.7 mmol) and diisopropylethylamine (DIPEA, 1.7 mL, 9.7 mmol) in order at room temperature. 2.8 mmol), ammonium chloride (0.5 g, 9.7 mmol) and diisopropylethylamine (DIPEA, 1.7 mL, 9.7 mmol). The reaction system was purged with nitrogen (N2) and then the reaction was stirred at room temperature for 6 hours. After completion of the reaction, the mixture was poured into water and extracted with ethyl acetate (EtOAc, 3 × 50 mL). The organic phases were combined, washed with saturated saline (2 × 20 mL), dried over anhydrous magnesium sulfate (MgSO4), filtered and concentrated under reduced pressure. The residue was treated with dichloromethane (DCM, 10 mL) to precipitate, and the precipitate was collected by filtration and dried to afford 2-amino-3-bromobenzamide as a pale pink solid (0.42 g, 84% yield). Liquid chromatography-mass spectrometry (LCMS, ESI) m/z 215/217 ([M+H]+).
