Uses

N-[4-(Aminosulfonyl)phenyl]-4-(3-phenyl-2-propenyl)-1-piperazinecarbothioamide is a a 4-thioureido-benzenesulfonamide derivative and a Wnt/β-catenin signaling inhibitor. Wnt/β-catenin signaling is involved with embryogenesis and tissue homeostasis.

Biological Activity

LF3 is a potent inhibitor of Wnt/β-catenin signaling th at suppresses cancer cells motility, cell-cycle progression, and the overexpression of Wnt target genes. LF3 is a potent and specific antagonist of the interaction between β-catenin and the transcription factor TCF4. LF3 blocks self-renewal of cancer stem cells.

in vivo

LF3 reduces tumor growth and induces differentiation in a mouse xenograft model of colon cancer. Tumor growth is significantly reduced when mice with GFPhigh cells are treated with LF3 at 50 mg/kg. LF3 treatment does not disturb the normal histology of the gut of mice[1].
