Example 32 Preparation of Intermediate Compound 32A: 3-Bromopyruvic acid (16.37 g, 98.05 mmol) was dissolved in anhydrous dioxane (90 mL), followed by the addition of ethyl thiooxalate (13.08 g, 98.22 mmol). The reaction mixture was stirred at 50 °C for 1.2 h, followed by concentration under reduced pressure at the same temperature to give a yellow solid crude product. The crude product was dissolved in a mixture of saturated aqueous sodium bicarbonate solution (150 mL) and deionized water (150 mL). The aqueous phase was extracted with ethyl acetate (6 x 400 mL). The aqueous phase was subsequently acidified to pH 2 with concentrated aqueous hydrochloric acid (21 mL) and precipitation was observed. The suspension was further extracted with ethyl acetate (5 x 500 mL). All organic phases were combined, dried over anhydrous sodium sulfate, filtered and concentrated under reduced pressure and dried under vacuum for about 15 h to afford compound 32A as a reddish brown solid (14.36 g, 73% yield), which could be used in subsequent reactions without further purification.
