GENERAL METHOD: Ethyl bromoacetate (374 mg, 2.2 mmol, 1.0 eq.) was slowly added dropwise to a solution of dichloromethane (10 mL) containing 4-methoxybenzylamine (307 mg, 2.2 mmol) and triethylamine (227 mg, 2.2 mmol, 1.0 eq.), and the reaction was carried out at 0°C. The reaction was carried out by stirring for 3.0 hours at room temperature. The reaction mixture was then stirred at room temperature for 3.0 h. The reaction was quenched with water upon completion. The organic layer was separated, washed with water and dried over anhydrous sodium sulfate. The organic solvent was evaporated under reduced pressure and the residue was purified by silica gel column chromatography using hexane/ethyl acetate as eluent. The target product ethyl 2-[(4-methoxybenzyl)amino]acetate was finally obtained in 42% yield. The structure of the product was confirmed by 1H-NMR (500 MHz, CDCl3): δ 1.21 (t, 3H, J = 7.1 Hz, -CH2CH3), 1.87 (s, 1H, -NH-), 3.33 (s, 2H, -CH2-), 3.68 (s, 2H, -CH2-), 3.72 (s, 3H, CH3O-), 4.13 (q , 2H, J = 7.1 Hz, -CH2CH3), 6.80 (d, 2H, J = 8.3 Hz, Ar-H), 7.19 (d, 2H, J = 8.3 Hz, Ar-H).
