Uses
Tasisulam is a anticancer agent and induces apoptosis via the intrinsic pathway, resulting in cytochrome c release and caspase-dependent cell death. Tasisulam inhibits mitotic progression and induces vascular normalization[1].
Synthesis
Acylsulfonamide 6 (18.335 kg, 44.17 mol), isopropanol (185 L), 51% NaOH aqueous solution (3.350 kg, 42.71 mol, 0.967 eq.) and deionized water (2.215 kg) were combined under N2. The slurry was stirred at 25C until it became a clarified solution. The solution was passed through a carbon filter, then filtered through a 0.45 m filter, followed by a 0.22 m filter and collected in a crystallization vessel, rinsed the initial vessel and filtered with IPA (90 L). The filtered solution was distilled at atmospheric pressure until 55L of the solution was retained in the crystallization vessel. The concentrated solution was cooled to 25C, inoculated with acylsulfonamide sodium salt 7 (0.178 kg) and stirred for at least 30 minutes. The product was crystallized by adding n-heptane (127.5 L). The resulting slurry was stirred at 25C for at least 30 minutes. The diluted slurry was thermally cycled three times from 25C to 60C at a rate of 0.5C/min. After maturation, the slurry was cooled to -5C at 0.5C/min and stirred for at least 1 hour, then filtered. The wet cake was washed with a 95:5 n-heptane/IPA mixture (92.5 L of n-heptane, 5 L of IPA) pre-cooled to -5C. Finally, the wet filter cake was dried in vacuo at 40 C by blowing with N2. This gave acylsulfonamide sodium salt product 7 (16.838 kg, 38.52 mol, 87.2% yield) which was white in color. (5-Bromothiophen-2-ylsulfonyl)(2,4-dichlorobenzoyl)amide sodium salt (Tasisulam sodium salt, 16.838 kg, 38.52 mol, 87.2% yield).
