The general procedure for the synthesis of (S)-2-amino-3,3,3-trifluoropropan-1-ol hydrochloride from the compound (CAS: 1255946-08-7) was as follows: (S)-3,3,3-trifluoro-2-((R)-2-hydroxy-1-phenylethyl)amino)propan-1-ol (28.0 g, 112 mmol) was dissolved in ethanol (300 mL). A 1.25 N methanolic hydrochloric acid solution (118 mL) and a 4 N dioxane hydrochloric acid solution (28.4 mL) were added sequentially. Subsequently, 20% w/w palladium hydroxide/carbon catalyst (3.86 g) was added to the reaction system and the reaction mixture was stirred under hydrogen atmosphere for 6.5 hours. After completion of the reaction, the catalyst was removed by filtration and the filtrate was concentrated under vacuum. The residue was ground with ether to give 17.1 g (92% yield) of (S)-2-amino-3,3,3-trifluoropropan-1-ol hydrochloride as a white solid. Its rotundity was [α]D -9° (c = 2.0, ethanol); 1H NMR (400 MHz, MeOD-d4) δ 4.19-4.11 (m, 1H), 3.96 (dd, J = 12.3, 4.3 Hz, 1H), 3.89 (dd, J = 12.1, 5.9 Hz, 1H).
