General procedure for the synthesis of 6-trifluoromethoxyindole from compound (CAS:1192478-00-4): aqueous potassium hydroxide solution (2 mL) was added to a methanol (2 mL) solution of compound 147 (ca. 200 mg, 0.673 mmol) at room temperature with continuous stirring of the reaction mixture for 16 hours. Upon completion of the reaction, water (50 mL) was added for dilution, followed by extraction with ethyl acetate (3 x 50 mL) to separate the product. The organic phases were combined, dried with anhydrous sodium sulfate and concentrated under reduced pressure to give the crude product. The crude product was purified by fast column chromatography with a gradient elution of hexane/ethyl acetate to pure ethyl acetate as eluent of 10:1 to give the final compound 148 (40 mg, 0.199 mmol, about 30% yield).
