The specific steps are as follows: Mix 3-(4-Bromophenyl)piperidin-2-one (7.6g, 30mmol) in... In deuterated methanol (100 g), under nitrogen protection, the mixture was heated to 40–45 °C, and sodium methoxide (3.2 g, 60 mmol) was added. The reaction was continued at 40–45 °C for 16 h. LC-MS analysis showed a deuteration rate of 90%. The reaction solution was cooled to 20–25 °C, quenched with 100 mL of ammonium chloride solution, extracted with ethyl acetate (100 mL × 2), dried, concentrated to obtain a crude product, and filtered with petroleum ether to obtain 7.0 g of a white solid, 3-(4-bromophenyl)-3-deuterated piperidin-2-one, with a yield of 92%. LC-MS analysis showed a deuteration rate of 92%, and NMR analysis showed a deuteration rate of 94%.
