The general procedure for the synthesis of (S)-3,3-difluoro-5-(hydroxymethyl)pyrrolidin-2-one from the compound (CAS:255903-83-4) was as follows: compound 10 (1.65 g, 8.37 mmol) was dissolved in a solvent mixture of acetic acid, acetonitrile, and water (14:3:3, v/v, total volume of 20 mL), and stirred until complete dissolution. Subsequently, the reaction mixture was heated and refluxed at 90 °C for 14 hours. Upon completion of the reaction, the solvent was removed by rotary evaporator to obtain the crude product. The crude product was purified by fast column chromatography (eluent 10% methanol/dichloromethane) to afford pure (S)-3,3-difluoro-5-(hydroxymethyl)pyrrolidin-2-one (Compound 11) as a white solid (1.04 g, 80% yield). The synthesis of compound 11 has been reported in the literature.
