6,7-Dichloro-1,4-dihydroquinoxaline-2,3-dione is a potential antagonist of the glycine site on N-??methyl-??D-??aspartate (NMDA) receptors and can be useful in the study of structure-??activity relationships of quinoxaline derivatives as Gly??/NMDA receptor antagonists.

Synthesis

Step 1: Acetic anhydride (10 g, 98 mmol) was added dropwise to a solution of 3,4-dichloroaniline (10 g, 93.5 mmol) and triethylamine (10.4 g, 103 mmol) in 100 mL of anhydrous dichloromethane at 0 ??C. The mixture was stirred at room temperature for 6 hours and then concentrated. The residue was dissolved in ethyl acetate, washed with water and brine, dried, concentrated, and purified by recrystallization.
