General procedure for the synthesis of 4-bromo-2,7-naphthyridin-1-amine from 2,7-naphthyridin-1-amine: 32 g of 2,7-naphthyridin-1-amine was dissolved in 200 ml of acetic acid at room temperature. Subsequently, 35 g of bromine was slowly added dropwise to 200 ml of acetic acid solution, controlling the reaction temperature not to exceed 25°C. After the dropwise addition, stirring of the reaction mixture was continued for 60 min. The reaction solution was diluted by pouring into 500 ml of water and the pH was adjusted to 7-8 with 500 ml of 25% ammonia solution. after stirring for 14 h, the brown precipitate was collected by filtration, washed with water and dried to give 28.3 g of crude product. The crude product was purified by fast chromatography using ethyl acetate/methanol as eluent, and finally 18.5 g of 4-bromo-2,7-naphthyridin-1-amine was obtained with molecular weight of 224.06 g/mol, and the [M+H]+ peak was detected by mass spectrometry, and the measured value was 224.
