Application
(R)-2-(dimethylaminomethyl)pyrrolidine hydrochloride can be used as an organic synthesis intermediate and a pharmaceutical intermediate, mainly in laboratory research and development processes and chemical production processes.
Synthesis
Take 1g of N,N-dimethyl-5-oxo-2-pyrrolidinecarboxamide, 0.92g of sodium borohydride, put into a 100mL four-necked flask, ice salt bath to lower the temperature, add 30mL of tetrahydrofuran, pass through nitrogen to protect, dropwise addition of 2.08g of iodine solution of tetrahydrofuran, after the completion of the temperature rise reflux. Thin-layer chromatography to detect the progress of the reaction, the end of the reaction, stop heating, the system was transferred to an ice bath, add water to destroy the sodium borohydride not involved in the reaction, filtration, filtrate extracted with ethyl acetate, combined with ethyl acetate, saturated sodium chloride to wash the organic layer, drying spin-drying, to obtain a white solid, i.e., (S)-1-benzyl-3,4-dihydroxy pyrrolidinium-borane complex. The white solid was dissolved in methanol and refluxed for 8h, spun dry, 10mL of ethyl acetate, 10mL of dilute hydrochloric acid (0.1N) was added, the aqueous layer was separated, the product was neutralized with sodium hydroxide to pH 8, the product was extracted from ethyl acetate, concentrated under reduced pressure to obtain an oily liquid, and acetone was recrystallized to give an off-white solid (R)-2-(dimethylaminomethyl)pyrrolidine hydrochloride.
