Application

3-Bromo-1-methylpyrrolidine is a colorless liquid and an important pharmaceutical intermediate. It can be used to prepare mGluR5 receptor allosteric modulators.

Synthesis

3-Hydroxy-1-methylpyrrolidine (5 g) was added in a 100 ml single-necked flask, n-hexane (50 ml) was cooled in an ice bath for 10 min, phosphorus tribromide (16.9 g) was added under cooling, and the temperature was raised to reflux for 2 h. Cooling was carried out. Neutralized to pH>8 by adding 25% KOH. extracted with ether. Combine the organic phases, add 1N hydrochloric acid 50ml, extract. Retain the aqueous phase, adjust pH>8, ether extract. Air dry. Obtained 3-bromo-1-methylpyrrolidine (1.1g).
