Description
Filgotinib Maleate, the maleate form of filgotinib, is an oral bioactive inhibitor of the tyrosine kinase Janus kinase 1 (JAK1) with potential anti-inflammatory and immunomodulatory activities. JAK1 is a key enzyme in cytokine and growth factor-mediated signaling pathways for inflammation and immune function. Therefore, inhibiting the activity of JAK1 helps to exert therapeutic effects in various autoimmune diseases, inflammatory diseases, and certain malignant tumors.

Mechanism of action
Filgotinib Maleate exerts its therapeutic effect by selectively inhibiting the JAK1 enzyme. By blocking JAK1, Filgotinib Maleate can effectively disrupt the signal cascade that leads to the production of pro-inflammatory cytokines. This inhibition leads to a significant reduction in inflammation and immune-mediated damage, thereby alleviating symptoms associated with autoimmune diseases such as rheumatoid arthritis.
Clinical Use
Filgotinib Maleate is a small molecule drug and has been approved for the treatment of rheumatoid arthritis (RA). It can also be used to treat Crohn's disease. Studies have found that it also has potential benefits in the treatment of other diseases such as ulcerative colitis, ankylosing spondylitis and psoriatic arthritis.
Side effects
Common adverse reactions of filgotinib maleate include upper respiratory tract infection, nausea, headache and elevated liver enzymes. Serious side effects may include infections (such as tuberculosis and shingles), thrombosis and gastrointestinal perforation. Therefore, filgotinib maleate is contraindicated in patients with active infection, severe liver function impairment, or a history of thrombotic events. It is also recommended that patients be screened for latent tuberculosis and vaccinated against shingles before starting treatment. If symptoms such as persistent fever, unexplained pain or shortness of breath occur, medical attention should be sought immediately.
