General procedure: pre-step A Synthesis of 2-fluoro-4-pyrimidinamine A mixed solution of 2,4-difluoropyrimidine (200 mg, 1.723 mmol) with ammonia (1600 μL, 22.18 mmol) in 1,4-dioxane (2 mL) was stirred at -21°C (salt-ice bath) for 5 min. Subsequently, methanol (1 mL) was added to the reaction mixture and the crude product was purified by reversed-phase high-performance liquid chromatography (RP-HPLC) to afford 2-fluoro-4-pyrimidinamine (94 mg, 0.831 mmol, 48% yield) as a white solid. Mass spectrum (electrospray positive ion mode) m/z 114.1 ([M+H]+).
