Synthesis

General method: Dihydroxypyridine 2 (0.05 mol, 1 eq.), tetramethylammonium chloride (11.0 g, 0.10 mol, 2 eq.), toluene (100 mL), and phosphorous trichloride (23 mL, 0.25 mol, 5 eq.) were added sequentially to an autoclave. The reaction kettle was sealed and the reaction was magnetically stirred at 200 °C for 5 hours. After the reaction was completed, the reactor was cooled to room temperature and opened carefully after the pressure was reduced to atmospheric pressure. The reaction mixture was slowly poured into warm water (80 mL) and diluted with ethyl acetate (100 mL). The organic and aqueous phases were separated and the organic phase was washed with brine (50 mL). The aqueous phase was back-extracted with ethyl acetate (2 x 80 mL). All organic phases were combined, dried with anhydrous sodium sulfate, filtered and concentrated under reduced pressure. The residue was purified by column chromatography to afford the target product 2,6-dichloro-5-phenylcyanopyridine as a white solid.
