ChEBI: An alpha-D-mannoside having 4-aminophenyl as the anomeric substituent.
Synthesis
(1) 1,2,3,4,6-penta-O-acetyl--D-mannose (12.82 mmol), p-nitrophenol (51.28 mmol), and zirconium tetrachloride (1.03 mmol) obtained from Preparation Example 1 were added sequentially to a 100 mL round-bottomed flask at 120 C, reduced to 0.08 MPa, stirred and melted, and after 1.3 h, TLC After 1.3h, the reaction was detected completely. The above reaction solution was taken and directly separated by column chromatography (V(petroleum ether):V(ethyl acetate) = 3:1). 4.93 g of p-nitrophenyl-2,3,4,6-tetra-O-acetyl--D-mannopyranoside was obtained in 81.9% yield. The product was characterized by 1H-NMR, spin, and melting point, and confirmed to be the target product.
(2) At room temperature, p-nitrophenyl-2,3,4,6-tetra-O-acetyl--D-mannopyranoside (10.51 mmol) and 40 mL of anhydrous methanol prepared in step (1) were added to a 100 mL round-bottomed flask, 10% (w/w) sodium methanol/methanol solution was added dropwise to adjust the pH value to be about 10, and the reaction was stirred for 5h, and the TLC The reaction was detected as complete, the pH was adjusted to 7 by adding an appropriate amount of cationic resin, filtered, and the filtrate was concentrated and separated by column chromatography (V(ethyl acetate):V(methanol) = 10:1) to obtain 2.63 g of 4-aminophenyl-ALPHA-D-mannopyranoside in 83.1% yield. The product was characterized by 1H-NMR, spin, and melting point, and confirmed to be the target product.
