ADRIAMYCINOL Usage And Synthesis

Uses

A metabolite of Doxorubicin as neoplasm inhibitor.

Definition

ChEBI: Doxorubicinol is a member of the class of tetracenequinones that is the major metabolite of the anthracycline doxorubicin, a chemotherapeutic agent effective against a broad range of malignant neoplasms. It is thought to exhibit cardiotoxic properties. It has a role as a drug metabolite and a cardiotoxic agent. It is a member of p-quinones, an anthracycline antibiotic, a deoxy hexoside, an aminoglycoside, a member of tetracenequinones, a member of phenols, an aromatic ether and a polyol. It derives from a hydride of a tetracene.

Synthesis

Adriamycin fermentation broth 2000 liters, add 1N of hydrochloric acid adjusted pH 1.0 for acidification, acidification 3 hours, plate and frame filtration, to get the pre-purified solution 1650 liters, HPLC detection, containing 1000g of adriamycin, chromatographic content of 16%. The pre-purified solution was adsorbed with 100 liters of HP20 resin, and after adsorption, 300 liters of 10% (V/V) aqueous ethanol solution with hydrochloric acid adjusting the pH to 2.0 as the pre-wash solution was used for pre-washing, and then 400 liters of 50% (V/V) aqueous ethanol solution with hydrochloric acid adjusting the pH to 2.0 as the eluent was used for elution and 300 liters of qualified eluent was collected. The resulting eluate contained 800 g of adriamycin with a chromatographic content of 79%.
