biological activity

1,4-DPCA, a potent and selective inhibitor of prolyl-4-hydroxylase, is also an inhibitor of collagen hydroxylation in chemicalbook cells of human foreskin fibroblasts with IC50 of 2.4µM. 1,4-DPCA inhibits the alpha isoform of prolyl-4-hydroxylase and stabilizes HIF-1alpha protein.

Description

1,4-DPCA (331830-20-7) is a potent inhibitor of prolyl 4-hydroxylase (IC50=2-4 μM).1 Inhibits collagen deposition via inhibition of collagen proline hydroxylation.2,3 Subcutaneous injection of a 1,4-DPCA containing hydrogel into Swiss Webster mice that do not show a regenerative phenotype increased expression of HIF1α over 5 days. Multiple injections led to regenerative wound healing after ear hole punch injury.4

Uses

1,4-DPCA nis a potent competitive inhibitor of avian prolyl 4-hydroxylase in embryonic chick tendon cells and human foreskin fibroblasts in vitro. It is a promotor of wound healing and tissue regeneration in mice.

in vivo

1,4-DPCA treatment suppresses connective tissue ingrowth in porous porous poly (lactic-co-glycolic acid) (PLGA) discs implanted in the peritoneal cavity for 28 days in mice. 1,4-DPCA is found to be effective at inhibiting collagen deposition within and on the outer surface of the disc, and also limited connective tissue ingrowth, but not to the extent of glucocorticoid inhibition[2].
