Uses

2-Amino-2-(3,4-dichlorophenyl)acetic acid hydrochloride is a pharmaceutical intermediate, and there are literature reports that it can be used to prepare MK2 inhibitors. MK2 is a key regulator of various pro-inflammatory mediators (including TNFα, IL-1β, IL-6, IL-8) and other important pathogenic signals in chronic inflammation, autoimmune diseases, and cancer.

Synthesis

Step 1.

Thionyl chloride (0.49 mL, 2M solution of CH2Cl2) was added to 2-amino-(3,4-dichlorophenyl)acetic acid hydrochloride (0.25 g, 0.97 mmol) in MeOH (13 mL). The mixture was refluxed overnight, cooled, and then concentrated in vacuum. The solid was ground with dichloromethane and filtered. To the resulting methyl ester was added a methanol (10 mL) solution in ammonia. after 24 hours, the mixture was concentrated to give a solid comprising 2-amino-2-(3,4-dichloro-phenyl)-acetamide hydrochloride and ammonium chloride.

Step 2.

A portion of this intermediate (75 mg) is added to 3-ethoxy-4-(pyridin-4-ylamino)-cyclobut-3-ene-1,2-dione (64 ) mg, 280 mol) in a solution of EtOH (4 mL). The reaction mixture was stirred at 60 C for 18 h and then concentrated in vacuum. Purification by HPLC (C18; 10 ) was performed. -100% gradient, H2O/ACN having 0.1% TFA) to obtain 2-amino-2-(3,4-dichlorophenyl)acetic acid hydrochloride.
