Uses

A very potent and selective inhibitor of dopamine uptake

Definition

ChEBI: A hydrochloride salt that is obtained by reaction of 1-[2-(benzhydryloxy)ethyl]-4-(3-phenylpropyl)piperazine with two equivalents of hydrogen chloride. Potent and selective inhibitor of dopamine uptake (KD = 5.5 nM in rat striatal memb anes).

Biochem/physiol Actions

GBR 12935 inhibits the dopamine and norepinephrine transporters with Kis of 21.5 nM and 225 nM, respectively. It does not inhibit the serotonin transporter (Ki = 6.5 mM). GBR 12935 binds to a nondopaminergic piperazine site in blood platelets and brain that has been identified as cytochrome P450.

in vivo

GBR 12935 dihydrochloride (1-32 mg/kg; repeat injection; 7 d) elevates locomotion activity to a greater extent in C57BL/6J mice than DBA/2J mice, and (10 mg/kg; injection; 7 d) results few mice sensitized to cocaine-induced stereotypy with repeated injections[3].

Animal Model:Adult male DBA/2J and C57BL/6J mice (22-30 g)[3]
Dosage:1.0, 3.2, 10, 32 mg/kg
Administration:Repeat injection; for 7 days
Result:Elevated locomotion activity to a greater extent in C57BL/6J mice than DBA/2J mice.
No stereotypy was induced by an eighth day challenge of 10 mg/kg GBR 12935 dihydrochloride in mice pretreated with seven dally injections of either 32 mg/kg cocaine or saline.
