To a suspension of potassium cyanide (1.90 g, 29.1 mmol) in methanol (35 mL) was added dropwise a methanolic solution (72 mL) of 3-bromo-1,1,1-trifluoropropan-2-oxime (5.00 g, 24.3 mmol) and the reaction was carried out at room temperature. The reaction mixture was stirred continuously for 3 hours at room temperature. Upon completion of the reaction, the solvent was removed by rotary evaporator and the residue was dissolved in ethyl acetate and stirred at room temperature. The resulting potassium bromide solid was removed by filtration and the filtrate was concentrated by rotary evaporation to give the crude product. The crude product was purified by silica gel column chromatography (Biotage: 25M, eluent 10% to 60% ethyl acetate/hexane, elution volume 550 mL). The pure fraction was collected and concentrated to afford the target product 3-(trifluoromethyl)isoxazol-5-amine (1.38 g, 37% yield). Mass spectrum (ESI) m/z: 153.0 ([M-H]-).
