Description

DBCO-PEG5-NHS Ester is a DBCO-containing PEG linker containing NHS ester that can react specifically and efficiently with primary amines (e.g. the side chain of lysine residues or aminosilane-coated surfaces) at neutral or slightly basic condition to form a covalent bond. PEG5 spacer increases the water solubility of compounds in aqueous media. DBCO is commonly used for copper-free Click Chemistry reaction.

Biological Activity

DBCO-PEG5-NHS ester is a PROTAC linker, belonging to the PEG class and the Alkyl/ether class, for the synthesis of a series of PROTAC molecules. DBCO-PEG5-NHS ester is a cleavable ADC linker for the synthesis of antibody drug conjugates (ADCs).

PROTACs contain two different ligands connected by a linker; one is a ligand for an E3 ubiquitin ligase and the other is for the target protein. PROTACs exploit the intracellular ubiquitin-proteasome system to selectively degrade target proteins.
ADCs are comprised of an antibody to which is attached an ADC cytotoxin through an ADC linker.
