The general procedure for the synthesis of tert-butyl 5-iodoisoindoline-2-carboxylate from di-tert-butyl dicarbonate and the compound (CAS:1221259-72-8) was as follows: the product of step A (0.77 g, 2.15 mmol) was dissolved in anhydrous pyridine (3 ml) with di-tert-butyl dicarbonate (Boc2O, 0.7 g, 3.2 mmol) and stirred at 70°C for The reaction was carried out for 1 hour. After completion of the reaction, the solvent was removed by distillation under reduced pressure. The residue was diluted to 20 ml with ether (Et2O) and filtered to remove insoluble matter. The filtrate was washed with dilute hydrochloric acid (5 ml), saturated sodium bicarbonate solution (5 ml) and brine, dried with anhydrous magnesium sulfate (MgSO4) and filtered. The filtrate was concentrated under reduced pressure and the resulting residue was purified by fast column chromatography (silica gel, dichloromethane:hexane=1:1) to afford tert-butyl 5-iodoisoindoline-2-carboxylate (0.58 g, 78% yield) as a milky-white oily substance.1H-NMR (CDCl3) data were as follows: δ 1.49 (s, 9H), 4.58-4.63 (m, 4H), 6.9- 7.04 (m, 1H), 7.5-7.63 (m, 2H).
