The general procedure for the synthesis of 3-bromo-2-hydroxy-5-iodopyridine from 3-bromo-2-hydroxypyridine is as follows: Example 142a Synthesis of 3-bromo-5-iodopyridin-2-ol In a 100 mL single-necked round-bottomed flask equipped with a magnetic stirrer, acetonitrile (50 mL), trifluoroacetic acid (TFA, 10 mL), 3-bromo-2-hydroxypyridine (4.0 g, 23.12 mmol), and N-iodosuccinimide (NIS, 5.2 g, 23.12 mmol) were added sequentially. The reaction mixture was stirred at room temperature for 15 hours. Upon completion of the reaction, the mixture was diluted with 100 mL of water and the resulting white solid was collected by filtration to afford the target product 3-bromo-5-iodopyridin-2-ol (142a, 6.6 g, 96% yield). Mass spectrometry analysis (MS-ESI): [M + H]+ m/z 300.
