JC-1 Usage And Synthesis

Description

JC-1 is a cationic carbocyanine dye that accumulates in the mitochondria of live cells in a potential-dependent manner.


The dye exists as a green-fluorescent monomer at depolarized membranes and low concentrations. At higher concentrations (aqueous solutions above 0.1 µM) and hyperpolarized membranes, the dye forms J-aggregates that exhibit an emission at the orange channel.


Healthy cells have high mitochondrial membrane potential, and the decrease of mitochondrial membrane potential is a marker of the early stage of apoptosis. All this allows the use of changes in the orange/green fluorescence ratio of JC-1 to determine healthy vs. depolarized mitochondria. The orange/green fluorescence ratio of JC-1 depends only on the mitochondrial membrane potential and not on other factors such as the size, shape, and density of mitochondria.

Uses

JC-1 is commonly used to determine mitochondrial membrane potential. It is a cationic carbocyanine dye that is membrane permeable and is often used for flow cytometry and fluorescent microscopy. General Description

A cationic, fluorescent, carbocyanine dye that can be used as a ratiometric indicator of mitochondrial potential ΔΨm in cells, tissues, and isolated mitochondria. It labels mitochondria with high membrane potential orange and mitochondria with low membrane potential green. Mitochondrial depolarization is indicated by a decrease in the red/green fluorescence intensity ratio. Ideal for detection of collapse of ΔΨm induced by uncouplers of mitochondrial respiration. The potential-sensitive color shift is due to concentration-dependent formation of red fluorescent J-aggregates. JC-1 has also been used in a functional assay of multidrug-resistance cells and in apoptosis-related mitochondrial modifications.
