Synthesis

General method: In step 2, the Schiff base intermediate was dissolved in 1,4-dioxane, iodine and potassium carbonate were added, and the cyclization reaction was carried out at 80 °C. The reaction mixture was refluxed for 4 hours and the progress of the reaction was monitored by thin layer chromatography (TLC). After completion of the reaction, the mixture was cooled to room temperature and the reaction was quenched by the addition of 5% sodium thiosulfate solution (20 ml). The target organic compounds in the reaction mixture were extracted with ethyl acetate. The organic phase was separated and collected and the solvent was removed by evaporation to give a solid product of 2-amino-5-(2,4-dichlorophenyl)-1,3,4-thiadiazole.
