Biological Activity

Sirtuin 1 (SIRT1) plays an important role in neuronal proliferation and vascular cells. It also plays an athero-protective role. Mutations in the SIRT1 gene affects the metabolism, cholesterol and f at deposition. SIRT1 has the ability to intermediate the insulin resistance through an inflammatory process, reactive oxygen species, gluconeogenesis and adiponectin level. It is an NAD(+)-dependent histone deacetylase involved in various nuclear events such as transcription, DNA replication, and DNA repair. SIRT1 protein binds and deacetylates the p53 protein. Expression of wild type SIRT1 in human cells reduces the transcriptional activity of p53 indicating th at SIRT1 is involved in the regulation of p53 function via deacetylation.

Description

SIRT1 Human Recombinant produced in E.coli is a single, non-glycosylated polypeptide chain containing 280 amino acids (254-495) and having a molecular mass of 31.6 kDa.
SIRT1 is fused to a 38 amino acid His-tag at N-terminus.

Background

Sirtuin-1 (SIRT1) belongs to the sirtuin family of proteins, homologs to the yeast Sir2 protein. Sirtuin family members are characterized by a sirtuin core domain and grouped into 4 classes. The functions of human sirtuins have not yet been established; though, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. It has been proposed that the human sirtuins may serve as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity.
