General procedure for the synthesis of 3-bromo-5-chlorothieno[3,2-b]pyridine from 5-chlorothieno[3,2-b]pyridine: 1.00 g (5.91 mmol) of 5-chlorothieno[3,2-b]pyridine, 497 mg (5.91 mmol) of sodium bicarbonate, 1.54 g (8.87 mmol) of potassium phosphate and 1.07 g (8.87 mmol) of magnesium sulfate were placed in a flask containing 50 mL chloroform. mmol) magnesium sulfate in a flask containing 50 mL of chloroform. The reaction mixture was stirred under reflux conditions for 30 minutes. Subsequently, 0.55 mL (10.82 mmol) of bromine was added and stirred overnight under reflux conditions. An additional 0.55 mL (10.82 mmol) of bromine was added and stirring was continued under reflux conditions for 24 hours. Upon completion of the reaction, the mixture was cooled to room temperature and extracted by adding water and dichloromethane. The organic layer was separated and the pH of the aqueous layer was adjusted to 14 with 5N NaOH and extracted with dichloromethane. All organic layers were combined and dried with anhydrous sodium sulfate followed by evaporation of the solvent. The crude product was purified by silica gel column chromatography (eluent: hexane:ethyl acetate = 5:1) to afford 796 mg (54% yield) of 3-bromo-5-chlorothieno[3,2-b]pyridine. The structure of the product was confirmed by 1H-NMR (CDCl3): δ 8.12 (d, 1H, J = 7.9 Hz), 7.81 (s, 1H), 7.36 (d, 1H, J = 7.9 Hz) ppm.
