General procedure for the synthesis of 2-amino-4-hydroxymethylthiazole from 1-bromo-3-hydroxypropan-2-one and thiourea: 1-bromo-3-hydroxypropan-2-one (60 g, 392 mmol) and thiourea (34 g, 451 mmol) were added to a 1 L round-bottomed flask containing 400 mL of ethanol. The reaction mixture was heated to reflux and held for 90 minutes. Upon completion of the reaction, the mixture was cooled and concentrated under reduced pressure to promote precipitate formation. The solid product (40 g, 80% yield) was collected by filtration and washed three times with hexane. The resulting product did not require further purification and could be used directly in subsequent reactions. The characteristic peak of the mass spectrum of the product was analyzed by LCMS (API-ES) at m/z 131.2 (100%, [M+H]+).
